HaloX™ Exchangeable Halotag Ligands Technology Launch

15 July 2023

Spirochrome & Abberior teamed up to bring the HaloX™ exchangeable halotag™ ligands technology1) available to the imaging community. Unlike regular Chloroalkane ligands, that react covalently with Halotag™ protein, HaloX™ ligands bind reversibly to Halotag™ protein. This unique feature allows, in addition to regular confocal microscopy, to perform STED, PAINT or MINFLUX superresolution imaging.

During STED, photobleaching occurs inevitably. The ability to exchange bleached fluorophores due to the non covalent nature of HaloX ligands extends the number of consecutive STED frames recorded.

HaloX™ ligands are based on bright and photostable dyes, including SiR, SPY555 or LIVE 590. The ligands are highly cell permeable and work with regular Halotag™ without the need for recloning or mutagenesis of previously established constructs or cell lines.
